A microfluidic platform for working with biological material

This bachelor's thesis focuses on the designing and fabrication of a microfluidic platform for cell lysis and subsequent analysis. This platform should be able to mechanically lyse cells in red blood suspensions efficiently and analyse the product using a two-electrode electrochemical system. The theoretical part of this thesis is dedicated to lab-on-a-chip technology and its components, such as microfluidic separators, mixers, lysis chambers and micro-reactors, their application and finally, their fabrication in clean rooms, along with basic physics of fluids in microchannels. The practical part of the thesis consists of designing three variants of the mentioned microfluidic platform, its fabrication in clean rooms and encapsulation using Parylene C polymer. The thesis follows the preparation of fabricated devices for testing and testing itself. The thesis concludes with imaging using a scanning electron microscope as a documentation of the fabrication process, resulting graphs from an electrode characterisation and images of cell behaviour within the lysis chamber.
Microfluidic device for cell counting and electrical lysis

This lecture presents two microfluidic devices as essential units for single cell proteomic analysis, firstly a device for cell counting based on resistive pulse sensing as an alternative to optical platforms based on visual control over the sample input, secondly a unit for electrical lysis of cells using direct current voltage pulses to lyse cells one by one with a possibility to analyze the distinct lysate zones by electrospray ionization mass spectrometry.
